AIM: To explore the clinical features, diagnosis and treatment experience and the distribution characteristics of pathogenic microorganisms of primary canaliculitis, and provide reference for its diagnosis and treatment. METHODS: Retrospective clinical study. A total of 119 cases(120 eyes)diagnosed as primary canaliculitis in the department of ophthalmology of Wuxi No.2 People's Hospital from June 2019 to February 2023 were included. The treatment methods were mainly divided into conservative treatment(removing canaliculus stones through lacrimal punctum combined with injecting antibiotic eye ointment into the tube)and surgical treatment. The inspection methods of pathogenic microorganisms included secretion smear microscopy and microbial culture.RESULTS: Primary canaliculitis was more common in middle-aged and older female, mainly manifested by long-term red eye and increased secretion; however, the majority was not accompanied by tearing. Totally, 118 cases(99.2%)had monocular disease, while 63 cases(63 eyes; 52.5%)had inferior lacrimal canaliculus disease. Laboratory examination: Among 119 cases(120 eyes), 4 cases(4 eyes)did not undergo laboratory examination, and the other 115 cases(116 eyes)were as follows: Gram staining microscopy of secretion smear showed that Actinomyces were detected in 102 cases(103 eyes; 88.8%), while no fungus was detected; Microbial culture: 85 cases(86 eyes; 74.1%)were positive for bacterial culture. A total of 111 bacterial strains were cultured, which contained 26 types of bacteria. Among them, 32 strains were aerobic(28.8%); 26 strains were anaerobic(23.4%); and 53 strains were facultative anaerobic(47.7%). The most common bacteria were streptococcus(20 strains), staphylococcus(13 strains), Propionibacterium(10 strains), and capnocytophaga(10 strains). Only 4 cases(4 eyes; 3.4%)of microbial cultures were positive for Actinomyces. Fungus was negative in all microbial cultures. Treatment: Of the 119 cases(120 eyes), 114 cases(115 eyes; 95.8%)were cured by conservative treatment of removing lacrimal canaliculus stones through lacrimal punctum and intracanalicular ointment infiltration(IOI), while 5 cases(5 eyes)were not effective in conservative treatment; however, all of them were cured after surgical treatment, and the cure rate for primary canaliculitis was 100.0%.CONCLUSION: The incidence of primary canaliculitis is low, and it is prevalent in middle-aged and older female. Single lacrimal canaliculus is more common, which could be missed and misdiagnosed in clinic. Actinomyces is the major pathogen observed mostly in mixed infections, with heterogeneous strains, mainly anaerobic or facultative anaerobic bacteria. Streptococcus and Staphylococcus are the most common whereas fungal canaliculitis is rare. The cure rate of primary canaliculitis is high after diagnosis, and IOI method is recommended as the initial treatment of canaliculitis.

RESUMO

Bilirubin has good anti-inflammatory, antioxidant and immunomodulatory effects, but its poor water solubility and low bioavailability greatly limit its clinical application. Researchers have developed bilirubin into various nanoparticles, which effectively eliminate the limitation of low solubility of bilirubin with the advantage of dosage form, so that they can maximize its pharmacological activities such as anti-inflammatory, anti-oxidation and immune regulation. Bilirubin nanoparticles have great application potential in a variety of gastrointestinal diseases, liver and kidney diseases, skin diseases, autoimmune diseases, islet transplantation and targeted therapy of tumors (both as a direct anti-tumor drug and as a drug delivery system). The study of bilirubin nanoparticles will promote the clinical application of bilirubin and the development of related new drugs.

OBJECTIVE To investigate the improvement effects of Arisaema Cum Bile on Parkinson’s disease (PD) model mice and its potential mechanism. METHODS Sixty male C57BL/6J mice were randomly divided into normal group, model group, Arisaema Cum Bile low-dose group [0.39 g/(kg·d)], Arisaema Cum Bile high-dose group [1.56 g/(kg·d)] and positive control drug Levodopa tablet group [80 mg/(kg·d)], with 12 mice in each group. Except that normal group was given constant volume of normal saline, other groups were given 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine [MPTP,35 mg/(kg·d)] intraperitoneally for 5 consecutive days to induce subacute PD model; after modeling, they were given relevant medicine continuously for 7 d; rod climbing test and line suspension test were performed 1 d before modeling, on the 5th day of modeling and after the last medication. The number of tyrosine hydroxylase (TH)-positive neurons in the substantia nigra of mice were measured by immunofluorescence; the levels of interleukin 1β (IL-1β) and tumor necrosis factor α( TNF-α) in serum and the levels of IL- E-mail:qhwang668@sina.com 1β, TNF-α, cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) in the substantia nigra of mice were measured by enzyme-linked immunosorbent assay. The expression levels of cAMP-dependent protein kinase catalytic subunit α (PKA C-α), glutathione peroxidase 4 (GPX4) and ferritin heavy chain polypeptide 1 (FTH1) proteins in the substantia nigra of mice was measured by Western blot. RESULTS After last medicine, compared with the normal group, mice in the model group had significantly longer pole-climbing time (P<0.01), significantly lower line suspension scores (P<0.01), significantly fewer TH-positive neurons in the substantia nigra (P<0.01), significantly higher serum concentrations of IL-1β and TNF-α and nigrostriatal concentrations of IL-1β, TNF-α, COX-2 and iNOS (P<0.01), while lower protein expression levels of GPX4, PKA C-α and FTH1 in the substantia nigra (P<0.05 or P<0.01). Compared with the model group, the above indexes of mice were significantly returned in Arisaema Cum Bile high-dose group (P<0.05 or P< 0.01). CONCLUSIONS Arisaema Cum Bile can improve motor impairment and reduce apoptosis of nigrostriatal TH neurons in MPTP-induced PD mice, and has neuroprotective effects on model mice; this may be related to its inhibition of neuroinflammation and the inhibition of ferroptosis by up-regulating PKA signaling pathway.

Epilepsy is a chronic brain disease characterized by seizures, and is one of the most common nervous system diseases in clinic practice with the recurrent, transient, and refractory characteristics. Clinically, western medicine therapy is mainly adopted in the treatment of epilepsy, but it is not conducive to long-term use for patients on account of severe side effects, which can result in abnormalities in the digestive system, central nervous system, hematopoietic system, urinary system, and liver function to varying degrees. Syndrome differentiation is usually used for the treatment of epilepsy by traditional Chinese medicine (TCM), which can avoid the side effects of western medicine treatment on the basis of improving patients' syndromes. The literature on TCM in the treatment of epilepsy in China and abroad indicates that the syndrome differentiation in TCM is often based on phlegm, blood stasis, wind, and deficiency, and the treatment methods include acupuncture, acupoint catgut embedding, moxibustion, Chinese medicine monomer, drug pair, and compound decoction. The various treatments of TCM play an important role in the comprehensive treatment of epilepsy through multiple channels and links, such as reducing the degree and number of seizures. This paper comprehensively summarized the clinical experience of TCM in the treatment of epilepsy, systematically expounded various treatment methods and ideas of TCM in the treatment of epilepsy, and deeply discussed the mechanism of TCM in the treatment of epilepsy, aiming to provide a theoretical basis for the clinical formulation of a reasonable individualized treatment plan for epilepsy and diversified ideas for the more effective treatment of epilepsy by TCM.

Traditional Chinese medicine has the characteristics of multiple components, pathways, and targets in the treatment of fracture healing, and has good therapeutic advantages and potential for fractures with complex pathological mechanisms. Based on this, the author summarized the mechanism of promoting fracture healing by the monomer components and compound formulas of traditional Chinese medicine and found that visfatin A, puerarin, and others can activate the mitogen-activated protein kinase (MAPK) signaling pathway; Xugudan, Guben zenggu formula and others can activate bone morphogenetic protein (BMP) signaling pathway; baicalin, Achyranthes bidentata polysaccharides and others can activate Wnt/β -catenin signaling pathway; apigenin, notoginsenoside and others can activate receptor activator of nuclear factor-κB (NF-κB)/receptor activator of NF-κB ligand/osteoprotegerin (RANK/RANKL/OPG) signaling pathway; Compound huoxue jiegu capsule, Jiangu granule and others can inhibit phosphoinositide 3-kinase/protein kinase B (PI3K/AKT) signaling pathway; icariin can activate Notch signaling pathway; Taohong siwu decoction, crocin and others can activate Hippo signaling pathway; jujuboside A and osthole can inhibit NF-κB signaling pathway, and thus promote fracture healing.

Pancreatic cancer is a disease with a high fatality rate, with a five-year survival rate of no more than 10% and a significantly increasing annual mortality rate. The common pathogenesis factors of pancreatic cancer are family inheritance, diet, pancreatitis, obesity, etc., among which, family inheritance of pancreatic cancer is the main reason, and about 7%-10% of patients have family inheritance. Surgery is an effective way to treat pancreatic cancer in patients and improve their survival, but most people are diagnosed with pancreatic cancer at intermediate and advanced stages and lose the opportunity for surgical treatment. Therefore, radiotherapy, interventional therapy, supportive treatment, immunotherapy, and other treatments are used clinically to relieve symptoms and prolong the survival of patients. The commonly used clinical drug is gemcitabine. Although it can inhibit tumor growth and improve the condition, it can bring side effects such as bone marrow suppression, rash, digestive tract side effects, and drug resistance. The damage of these side effects to the human body is systemic. Chinese medicine can be used alone or in combination with other treatment methods to reduce the toxic and side effects of chemotherapy, restore the physical energy of patients, and reduce its related complications. Chinese medicine contains a large number of active ingredients, including alkaloids, flavonoids, saponins, phenolic acids, and organic acids, with anti-inflammatory, anti-viral, anti-tumor, and other curative effects. Many clinical studies of traditional Chinese medicine (TCM) on cancers have verified that TCM plays a positive role in tumor prevention and treatment, especially in improving and controlling clinical symptoms, and also plays a good detoxification effect on radiotherapy and chemotherapy, with good results achieved in improving bone marrow suppression, improving immunity, improving quality of life, and prolonging survival. This paper reviewed the anti-pancreatic cancer mechanism of Chinese medicine monomers based on literature in China and abroad, aiming to provide new potential drug candidates for the treatment of pancreatic cancer.

ObjectiveTo explore the meridian tropism of components in Bupleuri Radix (Chaihu, CH) based on the model of nonalcoholic steatohepatitis (NASH) and clarify the substance basis of the meridian tropism of CH in Xiaoyaosan (XYS) by means of principal component analysis. MethodEighty SPF male C57BL/6 mice were randomly assigned into 8 groups, with 10 mice in each group. Except that the blank group was fed with the methionine choline-sufficient (MCS) diet, the other mice were fed with methionine choline-deficient (MCD) diet for 4 weeks to establish the nonalcoholic steatohepatitis (NASH) model. After the established model was confirmed by hematoxylin-eosin (HE) staining, the mice were administrated with corresponding drugs by gavage once a day for 4 weeks. Specifically, the 8 groups were XYS group (2.874 g·kg-1), XYS-CH group (2.445 g·kg-1), XYS-CH+volatile oils (Vol, 0.163 mg·kg-1) group, XYS-CH+polysaccharides (Pol, 24.067 mg·kg-1) group, XYS-CH+flavones (Fla, 2.241 mg·kg-1) group, and XYS-CH+saponins (Sap, 2.746 mg·kg-1) group. The model group and the blank group were administrated with the same volume of normal saline. After the last administration, the mice were sacrificed for the collection of blood and liver tissue. The pathological changes of liver were observed by HE staining and oil red O staining. Enzyme linked immunosorbent assay (ELISA) kits were used to determine the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), total cholesterol (TC), high-density lipoprotein (HDL), and low-density lipoprotein (LDL) in serum as well as malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) in liver. SPSS Statistics 23 was used for principal component analysis and comprehensive evaluation to determine the substance basis of the meridian tropism of CH in NASH mice. ResultCompared with the blank control group, the modeling led to hepatocyte swelling, increased fat vacuoles, and appearance of inflammatory cells. Further, the modeling elevated the levels of ALT, AST, TG, TC, and LDL and lowered the HDL level in serum, and it increased the MDA level and decreased the SOD, CAT, and GSH-Px levels in liver. Compared with the model group, the administration of XYS and XYS-CH in combination with the components of CH alleviated the oxidative damage in liver (P<0.05). The comprehensive score of the pharmacological efficacy was in a descending order as follows: XYS > XYS-CH+Sap > XYS-CH+Fla > XYS-CH+Pol > XYS-CH+Vol > XYS-CH. Among the chemical components of CH, Sap had the best effect. ConclusionSap lowers the blood lipid level, regulates the abnormal lipid metabolism, and alleviates the oxidative damage of liver, which is the substance basis for CH to exert the meridian tropism in liver.

Objective:To summarize the clinical manifestations and gene mutation features of patients with nucleotide excision repair (NER) disorders.Methods:A retrospective analysis was made on clinical data of patients with NER disorders who were admitted to the Chinese People′s Liberation Army General Hospital from October 2008 to February 2022 and diagnosed in the Outpatient Department of Beijing Children′s Hospital, Capital Medical University from October 2015 to February 2022.Literature on previously reported Chinese patients with NER disorders was reviewed.Results:(1)A total of 16 patients with NER disorders were enrolled, including 6 males and 10 females.The onset age was 7.5 (4.0, 12.0) months and the age at diagnosis was 42.0 (21.5, 77.0) months.There were 3 types of NER disorders: Cockayne syndrome (CS) in 13 cases, Xeroderma Pigmentosum (XP) in 2 cases and Cerebro-Oculo-Facio-Skeletal syndrome (COFS) in 1 case.Four disease-causing genes were detected: CSA gene in 11 cases, CSB gene in 3 cases, XPG gene in 1 case, and XPD gene in 1 case.The first symptoms of the 16 patients were photosensitivity and developmental delay, and neurological symptoms were observed in all the 3 NER disorder types.XP and CS patients had skin symptoms.CS patients presented typical facial features, visual and auditory impairment, microcephaly and changes in neuroimaging features.COFS patients showed intrauterine growth retardation.(2)Results of literature review: a total of 96 Chinese patients reported were retrieved, involving 6 disease types, including CS in 45 cases, XP in 44 cases, trichothiodystrophy in 4 cases, COFS in 1 case, XP-CS in 1 case, and ultraviolet sensitive syndrome in 1 case.Nine mutated genes were identified: CSA in 33 cases, XPA in 15 cases, CSB in 13 cases, XPV in 10 cases, XPC in 9 cases, XPG in 7 cases, XPD in 7 cases, XPF in 1 case, and MPLKIP in 1 case.The common symptoms were growth failure (62 cases), skin photosensitivity (61 cases), typical facial features (52 cases), mental retardation (49 cases) and microcephaly (48 cases). Among 36 cases had imaging data 33 cases(91.7%)had calcification of basal nucleus or globus pallidus.Three cases had intrauterine growth retardation and microcephaly during pregnancy. Conclusions:Patients with such prenatal manifestations as intrauterine growth retardation and microcephaly or with typical symptoms like skin photosensitivity, typical facial features, growth failure, mental retardation, hypertonia, and calcifications of basal ganglia should be suspected of NER disorders.Early genetic testing is recommended to confirm the diagnosis.

OBJECTIV E To establish the method for evaluating the quality o f Plantago asi atica and fried P. asiatica . METHODS The fingerprints of 15 batches of P. asiatica and 15 batches of fried P. asiatica were established by HPLC. The common peaks were identified with the Similarity Evaluation System for Chromatographic Fingerprinting of TCM (2012 edition), and similarity evaluation was performed. Analysis of chemical pattern recognition was performed by using SPSS 25.0 and SIMCA-P 14.1 software(cluster analysis ,principal component analysis and orthogonal partial least squares regression analysis ). The markers which affected the difference in the quality between P. asiatica and fried P. asiatica were screened with variable importance projection(VIP)value greater than 1. RESULTS There were 18 common peaks in the fingerprints of 15 batches of P. asiatica and 13 common peaks in the fingerprints of 15 batches of fried P. asiatica . A total of 8 common peaks were found in both of them. Their similarities were greater than 0.920. Two common peaks were identified as geniposidic acid ,acteoside. The results of cluster analysis showed that when the spacing was 10,the 30 batches of samples could be clustered into three categories ,with S 1-S5 as one,S16-S20 as one ,S6-S15 and S 21-S30 as one . The results of the pri ncipal component analysis showed that the cumulative variance contribution rate of the first two principal components was 82.575% . The results of the orthogonal partial least squares regression analysis showed that the VIP values of the three common peaks were greater than 1,namely peak E(acteoside), peak D (geniposidic acid ) and peak G. CONCLUSIONS Established fingerprints are stable ,simple sina.com and rapid. It can be used for the quality evaluation of P. asiatica and fried P. asiatica ,by combining with analysis of chemical pattern recognition. Acteoside ,geniposidic acid and the component represented by peak G may be the markers affecting the difference in quality of P. asiatica and fried P. asiatica .

Objective:To study minimally invasive treatment using percutaneous transhepatic choledochoscopic lithotripsy (PTCSL) to treat complicated hepatolithiasis.Methods:Patients with complicated hepatolithiasis who were treated at the Second Affiliated Hospital of Kunming Medical University from July 2017 to September 2021, were included into this study. There were 48 patients treated with PTCSL (the PTCSL group) and 150 patients treated with conventional open hepatectomy (the OH group). Propensity score matching (PSM) was used to match 40 patients in the PTCSL group with 40 patients in the OH group. There were 11 males and 29 females, aged 32 to 85 (59.3±12.7) years in the PTCSL group and 15 males and 25 females, aged 20 to 74 (55.4±10.9) years in the OH group. Theoperation time, intraoperative blood loss, hospitalization time, hospitalization cost, incidences of major complications (including pleural effusion, biliary bleeding), residual stone rates and recurrence rates were analyzed.Results:Baseline data were comparable between groups after PSM. For the PTCSL group, the operation time was (135.8±42.6) minutes, intraoperative bleeding volume 32.5(20.0, 50.0) ml, hospitalization time 13.5(11.0, 18.0) days, and hospitalization expense 3.4(2.9, 5.0) wanyuan compared with the corresponding figures in the OH group with operation time (350.7±113.8) minutes, intraoperative bleeding volume 475.0(200.0, 900.0) ml, hospitalization time 24.0(17.3, 28.8) days, and hospitalization expense 6.6(5.0, 8.9) wanyuan. The differences between the two groups were statistically significant (all P<0.05). The overall complication rate was 2.5%(1/40) in the PTCSL group and 22.5%(9/40) in the OH group (χ 2=7.31, P=0.007). The residual stone rate of 20.0%(8/40), stone recurrence rate of 17.5%(7/40), compared with the OH group with the stone residual rate of 27.5%(11/40), stone recurrence rate of 12.5%(5/40). There was no significant difference between the two groups in both the stone residual rates and stone recurrence rates (both P>0.05). Conclusion:In treatment of complex hepatolithiasis, the stone residual rate after repeated PTCSL was comparable to traditional open hepatectomy. PTCSL had the advantages of shorter operation time, less bleeding, lower hospitalization time, lower complication rate and lower hospitalization cost. PTCSL is worthy of wider application and popularization.

Objective:To investigate the effect of helicobacter pylori (Hp) infection and gastric polyps on colorectal cancer lesions.Methods:The clinical data of 2 034 patients with colorectal polyps and 118 patients with colorectal cancer detected by gastroscopy and colonoscopy examination in General Hospital of Beijing Jingmei Group from January 2020 to June 2021 were retrospectively analyzed. A total of 178 patients without colorectal polyps in the same period were treated as controls to analyze Hp infection and gastric polyps in patients with different ages, genders and pathological types.Results:The age and the proportion of male patients in colorectal cancer group were higher than those in the control group and colorectal polyps group (all P < 0.05). The Hp infection rates of the control group, colorectal polyps group and colorectal cancer group were 20.8% (37/178), 23.0% (467/2 034) and 27.1% (32/118), respectively, and the differences were statistically significant (all P > 0.05). Among 2 034 cases of colorectal polyps, there were 612 cases of inflammatory polyps, 371 cases of proliferative polyps and 1 051 cases of adenomatous polyps. The infection rates of Hp in the three kinds of colorectal polyps were 24.5% (150/612), 22.4% (83/371) and 22.3% (234/1 051), respectively. The incidence of patients with gastric polyps in the 3 groups was 34.6% (212/612), 38.3% (142/371) and 39.3% (413/1 051), respectively, and the differences were statistically significant (all P > 0.05). The age of the three colorectal polyps groups was significantly different from that of the control group and colorectal cancer group (all P < 0.05) except for inflammatory polyps and proliferative polyps groups. The proportion of female in three kinds of colorectal polyps combined with gastric polyps was higher than that in male patients (all P < 0.05). The age of Hp positive patients in the control group, different types of colorectal polyps groups and colorectal cancer group was lower than that in negative patients, but there was a statistically significant difference only in adenomatous polyps group ( P = 0.002). The age of patients with different types of colorectal polyps combined with gastric polyps was older than that of those without gastric polyps, and the age of patients with colorectal cancer combined with gastric polyps was younger than that of those without gastric polyps (all P < 0.05). There was no assocaiton of Hp infection and gastric polyps with colorectal polyps and colorectal cancer (all P > 0.05). Conclusions:Hp infection may promote the early occurrence of colorectal adenomatous polyps. There is no evidence that gastric polyps are associated with the risk of colorectal polyps. Female patients with colorectal polyps have a higher risk of gastric polyps.

Euryale f erox is the dry and mature seed kernel of Euryale ferox Salisb.,the effect of raw E. ferox is mainly to astringe spontaneous emission or leukorrhea ,while the effect of fried E. ferox is mainly to tonify the spleen and stomach. Therefore , processing has an important impact on the effect of E. ferox . The author summarizes the processing evolution ,chemical composition , pharmacological action and quality control of E. ferox by consulting past materia medica monographs and related research papers. The results show that the processing methods of E. ferox in the past include cleansing ,medicinal juice ,frying and steaming ;modern processing methods mainly continue to use cleansing and frying ,among which frying can be divided into stir-frying and bran-frying. E. ferox mainly contains polyphenols ,flavonoids,sterols and other components ,with antioxidant ,antibacterial,hypoglycemic and other pharmacological effects. At present ,scholars have established a variety of fingerprints to control the quality of E. ferox . Naringin,total amino acids and other components may be the differential components that affect the quality of E. ferox ,while the contents of heavy metals and sulfur dioxide are important indicators that affect the safety of E. ferox ,and α-tocopherol and gallic acid may be the quality markers of E. ferox . Later ,according to the basic properties of raw and processed products of E. ferox , pharmaceutical analysis methods can be used to comprehensively investigate the differences and change rules of chemical components in E. ferox before and after processing. The pharmacodynamic effects of E. ferox before and after processing can be evaluated by in vivo and in vitro models,so as to provide references for the inheritance of processing technology ,the formulation of processing standards and clinical application of E. ferox .

OBJECTIVE:To identify Panax notoginseng and its processed products . METHODS :The fingerprint was established by HPLC. Using ginsenoside Rb 1 as reference ,HPLC fingerprints of 15 batches of P. notoginseng and its processed products were drawn and the similarity evaluation was conducted by using the Similarity Evaluation System for TCM Chromatographic Fingerprints(2012 edition). The common peaks were confirmed by comparing with substance control. SPSS 21.0 and SIMCA 14.1 software were used to perform cluster analysis ,principal component analysis and orthogonal partial least squares-discriminant analysis;taking the variable importance projection (VIP)value greater than 1 as the standard ,the differential marker components causing the quality difference between P. notoginseng and its processed products were screened. IR fingerprints of P. notoginseng and its processed products were established by OMNIC 8.2.0 software,and the spectral similarity was evaluated ;double index sequence analysis was used to analyze absorption peaks of IR fingerprints of 15 batches of P. notoginseng and its processed products. RESULTS :There were 16 common peaks in the fingerprints of 15 batches of P. notoginseng , and the similarities were 0.911-1.000;there were 25 common peaks in the fingerprints of processed products ,and the similaritieswere 0.862-1.000. They had 12 identical common peaks ,and wang668@sina.com three of them were ident ified as sanchinoside R 1,ginsenoside Rg1 and ginsenoside Rb 1. Results of cluster analysi s showed that when the distance was 10,15 batches of P. notoginseng could be clustered into two categories ,SW1-SW5 into one category ,SH1-SH5 and SQ 1-SQ5 into one category ,ZW1-ZW5,ZH1-ZH5 and ZQ1-ZQ5 of 15 batches of processed products could be clustered into one category. When the distance was 5,15 batches of P. notoginseng could be clustered into three categories ,SW1-SW5 into one category ,SH2-SH5 and SQ 2 into one category ,SQ1, SQ3-SQ5 and SH 1 into one category. Fifteen batches of processed products could be clustered into two categories ,ZW1-ZW5 into one category ,ZH1-ZH5 and ZQ 1-ZQ5 into one category. The results of principal component analysis showed that the cumulative variance contribution rate of the first two principal components was 80.104% . The results of orthogonal partial least squares-discriminant analysis showed that the VIP values of the five peaks were greater than 1,which were peak H ,peak G ,peak J,peak F (ginsenoside Rg 1)and peak I. The similarity of IR fingerprints of 15 batches of P. notoginseng and its processed products were 0.889 7-1.000 0 and 0.972 8-1.000 0;the common peak rates were 80%-100%,and the variation peak rates were 0-17.65% and 0-18.75%,respectively. By comparing the wave numbers of absorption peaks ,it was found that there were differences between P. notoginseng at 3 440 and 1 450 cm-1 and processed products at 1 530 and 575 cm-1. CONCLUSIONS :Established HPLC fingerprint and IR fingerprint have good similarity ,and could effectively distinguish P. notoginseng and its processed products. P. notoginseng and its processed products from different habitats have high common peak rate and low variation rate ,and their chemical components are different ;peak H ,peak G ,peak J ,ginsenoside Rg 1 and peak I are differential marker components causing the quality difference between P. notoginseng and processed products.

OBJECTIVE:To isolate and purify the polysaccharides from Cucumis satiuus ,and to investigate its in vitro antioxidant activity. METHODS :The crude polysaccharides (CCL)were extracted from C. satiuus by water extraction and alcohol precipitation. CCL was separated and purified on Amberlite FPA 90Cl anion and Amberlite FPC 3500H cation exchange resin column and DEAE cellulose 52 column,so as to obtain 3 kinds of polysaccharides as CCL-M 2,CCL-M3,CCL-M6. The purity , molecular weight ,structure and monosaccharide composition of 3 kinds of polysaccharides were analyzed by HPLC-ELSD , UPLC-QTOF/MS,UV and IR. Using vitamin C (VC)as positive control ,the scavenging activity of CCL-M 2,CCL-M3 and CCL-M6(0.4,0.8,1.2,1.6,2.0 mg/mL)to 1,1-dibenzene-2-trinitrobenzene free radicals (DPPH·),hydroxyl free radicals (·OH) and superoxycyclic anion free radicals (O2-·)were investigated ,and IC50 was calculated. RESULTS :CCL-M2,CCL-M3 and CCL-M6 were homogeneous polysaccharides in the form of pyranose ,with molecular weights of 9.614×106,2.024×107 and 3.343× 107 Da,respectively. CCL-M 2 monosaccharides were composed of rhamnose ,semi-lactose aldehyde acid ,glucose, semi-lactose and arabic sugar ,with a molar ratio of 5.1∶2.6∶1.7∶1.4∶31.6. CCL-M 3 and CCL-M 6 monosaccharides were composed of mannose, rhamnose,glucose,semi-lactose,arabic sugar ,with a molar ratio of 1.7 ∶ 3.8 ∶ 6.4 ∶ 3.3 ∶ 1.3 and 1.3 ∶ 3.0 ∶ 3.2 ∶ 4.4 ∶ 8.9. The results of antioxidant activity in vitro showed that 3 kinds of homogeneous polysaccharides had certain scavenging activity on DPPH ·,·OH and O 2-·;the order of scavenging activity to DPPH ·was VC >CCL-M2>CCL-M3>CCL-M6(IC50 was 0.309, 1.240, 1.489 and 1.713 mg/mL); the order of scavenging activity to ·OH was VC >CCL-M2>CCL-M6>CCL-M3(IC50 was 0.968,1.032,1.233,1.356 mg/mL);the order of scavenging activity to O 2 -· was VC >CCL-M2>CCL-M3>CCL-M6 (IC50 was 0.335, 0.379, 0.812, 1.662 mg/mL). CONCLUSIONS:3 kinds of monosaccharides in the form of pyranose are isolated from C. satiuus ,and all of them have antioxidant activity in vitro ,and CCL-M 2 is the best.

There were no significant differences between the observation and control groups in both diseases about patient demographics and therapeutic approach. The CP implementation-quality differences between the two groups in both diseases were observed and compared. Significant differences were found between the two groups in both diseases about CP completion rate(97.2% versus 90.5% and 97.7% versus 90.1%), the rate of outside-CP doctor′s orders((4.6±1.3)% versus (19.3±5.3)%and(6.1±1.7)%versus (20.3±5.1)%), the hospitalized period((2.7±0.3)d versus (3.2±0.4)d and(5.6±0.4)d versus (6.2±0.5)d), the hospitalization costs and the satisfaction rate.@*Conclusions@#ACP belongs to one of compound CPs, is appropriate to use in these solitary diseases with significant variations. ACP can deal with the CP problem of significant variations rooted in the diseases or treatments, thus contributing to promotion and application of CP.

BACKGROUND:Previous research have confirmed that CD34 is closely related to oncogenesis, progress, recurrence, metastasis and drug-resistance of various cancers, but its role in nasopharyngeal carcinoma remains unclear. OBJECTIVE:Tosortcels positive and negative for CD34 in nasopharyngealcarcinoma cel lines and to detect cel proliferation and migration. METHODS:Expressionsof CD34 in nasopharyngeal carcinoma cel lines 5-8F, 6-10B, CNE1 and CNE2 were detected by flow cytometry. And CD34+and CD34-cels were sorted based on cel surfacemarkers for purity identification. Afterwards, proliferation and migrationof CD34+and CD34-celswere detected by MTT assay, colony-formation assay and scratch assay. RESULTS AND CONCLUSION:Al four nasopharyngeal carcinoma cel lines expressed CD34 in 0.1%-0.2%, and the level of CD34 was closely related to the cel growth density. The purity of CD34+cel was more than 98% in the sorted CD34+celpopulations, but no CD34+cels were found inthe sorted CD34-celpopulations.At 1, 3, 5 and 7 daystheproliferation rate of CD34+cel, populationswas significantly higher than that of CD34-cels (P< 0.05). Consistently, thecolony-formation efficiencyof CD34+cel was significantlyhigher than that ofCD34-cels (P< 0.05). Moreover, CD34+cels migrated significantly faster than CD34-cels by scratch assay (P< 0.05). In conclusion, CD34+cels culturedin vitro display higher proliferation and migration capacities, indicating that CD34+celshavethe potential of nasopharyngeal carcinoma stem cels.

ObjectiveTo observe and explore the effects ofJiakangning Capsules on the expression of ERK1/2 gene and proliferation of FRTL-5 by studying the effects ofJiakangning Capsules collaborated with siRNA on interfering ERK1/2 gene in FRTL-5.Methods FRTL-5 cells in good conditions were divided into control group, negative control group,Jiakangning Capsules collaborated with siRNA group, siRNA interference group, and Jiakangning Capsules group. RT-PCR was performed to detect the expression of ERK1/2 gene in mRNA levels in FRTL-5; Western blotting was performed to detect the expressions of ERK1/2 and p-ERK1/2; CCK8 method was used to detect cell proliferation.Results RT-PCR results showed that the ERK1/2 mRNA expression of the control group and negative control group were of no significant difference (P>0.05); compared with the control group, siRNA interference group andJiakangning Capsules group could inhibit ERK1/2 gene mRNA expression in FRTL-5 (P<0.01). Compared with the control group and negative control group, the ERK1/2 mRNA expression of Jiakangning Capsules collaborated with siRNA group decreased obviously (P<0.01). Compared with the control group and negative control group, the expression of p-ERK1/2, ERK1/2 ofJiakangning Capsules group, Jiakangning Capsules collaborated with siRNA group, and siRNA interference group decreased obviously (P<0.01). At the time of 24 h and 48 h after transfection, according to the results of CCK8, compared with the control group and negative control group, the cell proliferation ofJiakangning Capsules group, Jiakangning Capsules collaborated with siRNA group, and siRNA interference group was inhibited (P<0.01).Conclusion Jiakangning Capsules have blocking effect on the phosphorylation of ERK1/2. After ERK1/2 gene was silenced, the thyroid cell proliferation was inhibited. Jiakangning Capsules can collaborate with ERK1/2-siRNA to inhibit FRTL-5 cell proliferation.

Objective To investigate the effect of high mobility group box chromosomal protein 1 (HMGB1) knockdown on improving renal function and decreasing cell proliferation of glomeruli in lupus nephritis (LN) MRL/Faslpr mice.Methods Twenty-four MRL/Faslpr mice were randomly divided into 3 groups:LN model group,shHMGB1 group and empty plasmid group.Besides,eight MRL/MpJ mice,age and mass matched to the MRL/Faslpr mice,were chosen as normal control group (shNC group).Electroporation technology was used for in vivo transfection in treatment group.shHMGB1 group and empty plasmid group were transfected by electroporation technology for shHMGB1 plasmids and empty plasmid,LN model group and normal control group were transfected only with saline.Automatic biochemical analyzer was used to detect serum urea nitrogen (BUN) and creatinine (Scr) levels and 24 h urinary protein (UP) was tested.HE staining was used to detect the pathological change of renal tissues; real-time PCR,immunofluorence staining and Western blotting were used to detect the mRNA and protein expression of HMGB1 and PCNA.Results (1) The HMGB1 mRNA and protein expression in LN group increased compared with those in control group,HMGB1 mRNA and protein expression in shHMGB1 group reduced compared with those in LN model group (all P < 0.05).(2) 24 h UP of MRL/Faslpr mice in shHMGB1 group significantly reduced compared with those in LN group (P < 0.05).(3) Immunofluorence and Western blotting showed that positive signal of proliferating cell nuclear antigen (PCNA) was mainly located in nuclei,PCNA mRNA and protein in glomeruli of LN model group increased compared with those of control mice (P < 0.05).Interestingly,PCNA expression in glomeruli of shHMGB1 group remarkably reduced (P < 0.05).Conclusions shHMGB1 significantly improves renal function and decreases cell proliferation of glomeruli in LN MRL/Faslpr mice.

Objective To identify Isatidis Radix in different areas by SDS-PAGE;To provide the basis for the identification and quality evaluation of Isatidis Radix. Methods By using SDS-PAGE technology, the protein profiles of Isatidis Radix in Heilongjiang and Hebei regions were established. Results Isatidis Radix protein in Heilongjiang contained about 13 protein subunits, and the major subunits were 10.5 kD, 23.0 kD, 24.9 kD, and 44.1 kD. Isatidis Radix protein in Hebei contained about 12 subunits, and the major subunits were 55.6 kD, 45.9 kD, 34.3 kD, 18.9 kD, 12.4 kD, and 10.5 kD. Conclusion SDS-PAGE technology can be used as one of the references to identify Isatidis Radix in different areas.
